Set the background a colour you aren't using.
Go to Iaza.com
Use it to open up your image from your PC.
Select 'Transparent' and then click the color you want transparent (I think that's how it goes)
The image is ready.

OR

Use Paint.net and select the magic want
Set Tolerence to 0%
Click the delection you want to make transparent, and delete.
Save as a PNG.

Also, looking at your banner, make SURE you save as a PNG. For sprite works, JPG and JPEG badly effect your colours, whilst Bitmap (BMP iirc) takes up a lot of space on your PC.
